This drawing shows the mechanism of In vivo Confocal Microscopy
The In vivo Confocal Microscope can focus through the entire range of a normal human cornea from the epithelium to the endothelium.  This series of scans shows a) epithelium b) corneal nerves c) keratocytes d) deeper keratocytes e) endothelium and f) computer generated slice of cornea.
One of the outstanding uses of the In vivo confocal microscope is in the positive diagnosing of Acanthamoeba Keratitis.  The round white dots on this scan show the active acanthamoeba. 
This instrument can also pick up the acanthamoeba that have formed a cyst around themselves called a trophozoite.
