CYP17 (17-hydroxylase, left) and CYP21 (21-hydroxylase, right) share 29% amino acid identity yet selectively hydroxylate progesterone at carbon atoms that are only 1.6 Å apart.  Our laboratory uses computer modeling, molecular dynamics simulations, and site-directed mutagenesis experiments to study why these enzymes appear so similar yet possess different activities that fulfill diametrically opposite functions in human physiology.
