Photo-caged Fluorescent Molecules

Description: Photocaged fluorescent molecules are important tools for tracking molecular dynamics, especially high spatiotemporal resolution in biological systems.  This technology features a new class of caged coumarin fluorophores which displays more than 200-fold fluorescence enhancement after UV photolysis.  They have remarkable uncaging efficiency, approximately two orders of magnitude higher than previously described caged fluorophores.  Other significant features include a non-UV excitation wavelength at 408 nm and an emission of blue light which is spectrally complementary to many existing fluorescent sensors in the red and green regions.  An example of this technology has been successfully conjugated and tested in a number of cultured mammalian cells for in vivo imaging applications.  The combined advantages of robust fluorescence contrast enhancement, remarkable high uncaging cross sections, noninvasive cellular delivery, and flexible chemistry for bioconjugations should generate broad applications of these caged coumarins for biochemical and biomedical imaging.
