Description
Chromosome analysis of chorionic villus biopsy for the prenatal diagnosis of cytogenetic abnormalities.
Methodology
Chorionic villi are cleaned by physical and enzymatic methods.  Villi are minced, disaggregated, cultured in four to six vessels, and then harvested and G-banded with trypsin-Wright stain.  The chromosomes in twenty metaphase spreads from at least two separate cultures are counted, five spreads are fully analyzed and two are karyotyped.

Specimen Requirements

Approximately 25 mg of uncleaned specimen usually provides enough material for cytogenetic studies.  Up to 50 mg may be required if biochemical or DNA studies are also requested.  Place the chorionic villus sample into a tube containing 10 ml of sterile tissue culture medium. 

Note: Deliver to lab at room temperature within 24 hours. If collected over a weekend, call the laboratory to make arrangements for specimen delivery. May be refrigerated if not transported immediately. Do not freeze. Protect from heat or cold.

Frozen, fixed or grossly contaminated specimens.
