Description
These are a set of quantitative tests used to determine the absolute numbers of various cell types in the blood (e.g., total T cells, CD4 T cells, CD8 T cells, B cells, NK cells). The actual test is determined by the clinical situation. For example, in HIV disease, only a CD4 count is typically monitored. In a workup for immunodeficiency, the physician may request quantification of all subsets. The client provides the total leukocyte (WBC) count and the lymphocyte percentage ("CBC data"). The lab will determine the relative proportion of the requested cell subsets within the lymphocyte population. The absolute count is determined by a simple calculation, based on the CBC data.
Methodology
Flow Cytometry, Dual Platform
(CD3/CD4/CD8/CD45) and/or (CD3/CD16 CD56/CD45/CD19)
Performed
Monday - Friday.
Turnaround Time
Results available next business day.  Same day results are available if sample is received in the lab by 1:00 pm. Stat service for this testing is available with direct coordination with the laboratory staff (additional charges may apply).
Specimen Requirements

Special Instructions:

  • Please submit WBC count and Percent (%) Lymphs from CBC report from SAME date of collection.
  • SPECIMEN CANNOT BE FROZEN

Peripheral Blood:

  • One full 5 mL (2.5 mL minimum) lavender top tube (K2-K3EDTA)  is preferred, or
  • One full 3 mL ACD is an acceptable alternative anticoagulant, but not when used in Plasma Separator Tube (PST).

Transport Instructions:

  • Room temperature (20-25°C)

Specimen Stability

  • K2-K3EDTA specimens should be processed within 30 hours of collection.
  • ACD specimens should be processed within 72 hours of collection.

All tubes must be properly drawn; partial draw may cause cell lysis and loss due to hypertonic osmotic changes.
