Description
Cytogenetic analysis of cultured fibroblasts to assess for the presence of constitutional (congenital) chromosome aberrations.
Methodology
Tissue is minced and/or enzymatically disaggregated and is cultured until sufficient dividing cells are present (10 days to 4 weeks). Standard cytogenetic harvest. G-banding with trypsin-Wright stain. The chromosomes in twenty metaphase spreads are counted, five cells are fully analyzed and two are karyotyped.

2 - 3 weeks (dependent on growth rate of cultured cells).
Specimen Requirements

At least 5mm3 of aseptically collected fresh tissue from surgery or autopsy, or a generous 2 mm diameter (minimum) full thickness skin biopsy. Do not send skin from autopsy. Kidney, lung, fascia, cartilage and chorionic villi are preferred specimens from autopsy. Place specimen in 10 ml of sterile tissue culture medium and refrigerate until transported. Do not freeze. Do not put in formalin or other fixative.

Note: Deliver to lab at room temperature within 24 hours. If collected over a weekend, call the laboratory to make arrangements for specimen delivery. May be refrigerated if not transported immediately. Do not freeze. Protect from heat or cold.

Rejection Criteria

Frozen, fixed or grossly contaminated specimens. Specimens sent without tissue culture medium.
