Description
Cytogenetic analysis of unstimulated peripheral blood leukocytes to assess for the presence of a neoplastic clone.
Methodology
Cells are harvested from two or more unstimulated cultures. Twenty metaphase spreads are fully analyzed and two karyotypes are prepared. If abnormalities are found, at least two karyotypes are prepared for each abnormal clone.
Performed
Daily.
Turnaround Time
1 - 3 weeks. Preliminary results can be provided sooner if necessary.
Specimen Requirements

5 - 10 ml aseptically collected blood in a green top sodium heparin tube. Oncologic diagnosis (suspected or confirmed) must be provided.

Note:

  • Bone marrow aspirate is always the preferred specimen for cytogenetic analysis in cases of suspected leukemia. Blood may be substituted in cases of untreated chronic myeloid leukemia (CML) or in acute leukemias with WBCs >35,000, but the rate of abnormal clone detection is always better in bone marrow.
  • Deliver to lab at room temperature within 24 hours. If collected over a weekend, call the laboratory to make arrangements for specimen delivery. May be refrigerated if not transported immediately. Do not freeze. Protect from heat or cold.
Rejection Criteria
Frozen, clotted, hemolyzed or grossly contaminated specimens. Incorrect anticoagulant used.
